Abstract: This research work was performed on Coccinia indica Wight & Arn (Kundru; Ivy-gourd; Cucurbitaceae) for antioxidant and anti-hepatotoxic activity against paracetamol in wistar rats. Kundru contains indicated phenolics, flavanoids, glycosides, terpenoids and phytosterols as SPMs. Extractive values in leaves (ASEV:8.6%; WSEV: 24.8%; AIAV: 1.783%; WSAV: 8.93%) and fruits (ASEV:8.8%; WSEV: 25.20%; AIAV: 1.834%; WSAV: 9.68%) were calculated. Practical yield were 2.4% (leaves) and 2.72% (fruits) and Quercetin isolated (Rf value 0.46 and λmax 370 nm) from MECIL and MECIF. In anti-hepatotoxic activity, PCM induced hepatoxicity by glutathione depletion, necrosis of hepatocytes, elevated hepatic biochemical parameters with centilobular necrosis. MECIF and MECIL extract produced remarkable anti- hepatotoxic activity with significant alleviation of LFTs and reddish coloration, normal anatomical architecture. Anti-hepatotoxic effects were comparatively lesser then standard (Silymarin). and mechanism include anti-inflammatory (inhibition of phopholipase A2), membrane stabilization, anti-oxidant, glutathione restoration and enhanced RNA synthesis.
