Analytical Method Development and Validation for The Simultaneous Determination of Tadalafil and Tamsulosin Hydrochloride by UHPLC in Oral Solid Dosage Forms
Tamsulosin hydrochloride and Tadalafil belongs to a class of Alpha blockers drug used to treat Erectile dysfunction during sexual stimulation. The Ultra High Performance Liquid Chromatography method was used to develop and validate a new combination method of Tamsulosin-hcl and Tadalafil in a simple, accurate, rapid and simultaneous assay. The separation was done on an X-Bridge C18 column (4.6 × 50 mm: 3 microns) with a mobile phase of Ethyl Acetate: Toluene: Methanol: Ammonia (3:2:5:0.6) in isocratic mode and pumped according to flow rate. 0.5 ml/min and the compound was detected at 210 nm. Run time was 5 minutes. The retention time was 2.5 minutes and 3.1 minutes for tamsulosin hydrochloride and tadalafil. The % determination was 99.83% for Tam HCI and 100.78% for Tad. The retention time and run time were simple, fast and economical, which can be applied in regular quality control tests in the pharmaceutical industry.
